I do need help
Im just started learning labview
I dont know how to connect and program 6221
I connected the device(6221 connected to 2182a) via rs232
Can anybody explain step by step what should i do
I searched a lot but couldnt find any thing
for your information again
I havent download any drive or anything

If you are referring to NI USB-6221, it is a DAQ device and doesn't support RS-232 natively.
You can buy an off-the-shelf USB-to-RS232 converter and run Keithley 2182A Nanovoltmeter - Instrument Driver

Sorry, there was a mistake in my question that I realized now
two devices connected with rs 232
I wanted to program through digital io that i used rs232 to usb cable to connect my laptop

We do talk about the USB6221 from NI? Or do we talk about the Keithley6221?
I guess you talk about two Keithley devices (6221 and 2182): How exactly did you connect the Keithley6221 to your computer? One bullet point says "RS232", the other says "digital IO port"…
