Right. I checked the manual prior to posting and it is still not clear which wire pairings go to which pins. For example, I have 2 wires from a transducer, where do they go for Channel 0, Channel 24 etc?

From the manual:
NI 6031E, NI 6033E, and NI 6071E only
For these extended AI devices, the AI signals are AI <0..63>, AI SENSE, AI SENSE 2, and AI GND. In single-ended mode, signals connected to AI <0..63> are routed to the positive input of the PGIA. In differential mode, signals connected to AI <0..7, 16..23, 32..39, 48..55> are routed to the positive input of the PGIA, and signals connected to AI <8..15, 24..31, 40..47, 56..63> are routed to the negative input of the PGIA.
And by looking at the pinout of the card, channel 0 would use AI 0 (+) and AI 8 (-), which are pins 3 and 4 respectively. The SCB-100 should be a 1:1 direct wiring scheme, so use terminals 3 and 4. You may need bias resistors for DIFF mode, please look at the manual.
